I asked Caleb about this once, and apparently, the article's URL will have a number added to the end. For example, my newest article's URL is http://www.papertank.com/article/Currents, but if someone else submitted an article called Currents, their article's URL would be something like http://www.papertank.com/article/Currents2.
As far as I know, this has never happened, but it probably will when I submit my next story. My submission for the recent writing contest on Newgrounds was called Homecoming, and there's already an article here with that title. If I ever submit Homecoming to Papertank, we'll see if the system works.

Update:
I just submitted Homecoming, and it turns out the system tags a "_1" onto the end of the URL. (The original article by Soli is located at http://www.papertank.com/article/Homecoming, and mine is at http://www.papertank.com/article/Homecoming_1. If a third story with that title were submitted, it would be at http://www.papertank.com/article/Homecoming_2.)
As for taglines, they don't really matter. Two articles can have the same tagline without any problems. The URL only changes if two titles are identical.
